You MIGHT be able to, I honestly don't know and I don't have a cable here at my office to check. But... If you can plug your D3300 into your computer using the cable AND find the SD card listed as a drive on your computer, you can probably get away with it.
Thing is, to update your firmware you need to be able to put the .bin file on the SD card but
NOT in a folder on the SD card; and that's
crucial to the update happening. If you can do that on your PC, using the cable, you won't need the USB card reader. Even so, using the SD card reader will probably prove faster and more convenient than connecting your camera to your PC with the cable every time you want to transfer photos.
